  1. What’s an RBS and a rho independent termination sequence and what are the macromolecules involved.
  2. If you have a gene in an ORF of a second gene can you change the expression of the first gene without moving it.
  3. Describe the lac system with all its components in its biological form.
  4. The lac system is an all or nothing system, describe what that means. Design a simple experiment to showcase with E.coli with GFP controlled by Placz to show this all or nothing behavior. If you add glucose to your experiment does anything change?
  5. What other than all or nothing can you create with regulation and how would you modify the lac system to exhibit this behavior?
  6. Design a (biological) bandpass filter with E.coli that sends out AHL. It creates a band on a petri dish.
  7. What would you change to increase the distance to the inner circle?
  8. What would you change to decrease the distance to the outer circle?
  9. How do you design another bandpass with another fluorescent protein that is further away from the centre without the two bandpasses being able to overlap?
